Direct determination of ABO blood group genotypes from whole blood using PCR-amplification of specific alleles method

چکیده
ABO antigens are known to be oligosaccharide antigens, and are widely expressed on the membranes of red blood cells and tissue cells. Therefore, the determining ABO blood group antigens is important in both transfusion and organ transplantation, and is one of the markers used for personal identification in forensics. The ABO blood group is currently determined by the presence of A and B antigens on red blood cells using serological tests in clinical laboratories. The gene sequences of the ABO blood group in chromosome 9q34.1-q34.2 have also been determined. Accordingly, it has become possible to genetically analyze the ABO blood group using molecular biological techniques. We recently developed an ABO genotyping method based on PCR amplification of specific alleles (PASA) using DNA extracted from blood and saliva. However, the extraction and purification of DNA is necessary prior to PCR because blood and saliva contain various substances that inhibit PCR. Furthermore, the PCR amplification of specific alleles (PASA) method requires specificity and stability for allele-specific amplification. Therefore, it is very difficult to use whole blood directly. Here, we described the development and use of ABO genotyping from whole blood using a commercially available reagent kit, which can effectively neutralize inhibitory substances present in the blood. In this study, for all genes of the six major ABO genotypes and cisA2B3 genotype of the AB variant, only specific bands were clearly amplified, whereas non-specific bands were not amplified at all. In addition, this method was able to determine ABO genotyping using 5-fold diluted fresh whole blood, or 5-fold diluted whole blood that was freeze-stored in 100 μL aliquots at -20 C by subdivided for a maximum of 30 days. This analysis method to determine ABO blood group genotyping is simple and useful, and is expected to be used widely throughout research and clinical laboratories and forensic fields.

ABO glycosyltransferase genotyping by polymerase chain reaction using sequence-specific primers.
Serological typing for the classical ABO blood groups is routinely performed using anti-A and anti-B antisera of polyclonal or monoclonal origin, which are able to distinguish four phenotypes (A, B, AB, and O). Modern molecular biology methods offer the possibility of direct ABO genotyping without the need for family investigations. Typing can be done with small amounts of DNA and without detec...
متن کاملA rapid long PCR-direct sequencing analysis for ABO genotyping.
ABO is the most clinically important blood group system in transfusion and transplantation medicine. The popular ABO genotyping methods, such as the sequencing of exons 6 and 7 and sequence-specific primer (SSP)-PCR, often lead to ambiguous typing results. Long PCR-sequencing method was designed to analyze two regulatory regions (promoter and CBF/NF-Y enhancer regions) and all genomic sequences...

ABO genotyping is commonly used in several situations, including blood transfusion, personal identification and disease detection. The present study developed a novel method for ABO genotyping, using loop‑mediated isothermal amplification (LAMP). This method allows the simultaneous determination of six ABO genotypes under 40 min at a constant temperature of 62˚C. The genotypes of 101 blood samp...

BACKGROUND: The ABO blood groups result from DNA sequence variations, predominantly single nucleotide and insertion/deletion polymorphisms (SNPs and indels), in the ABO gene encoding a glycosyltransferase. The ABO blood groups A(1), A(2), B and O predominantly result from the wild type allele A1 and the major gene variants that are characterized by four diallelic markers (261G>del, 802G>A, 803G...
متن کاملMultiplex allele-specific amplification from whole blood for detecting multiple polymorphisms simultaneously.
Allele-specific amplification on the basis of polymerase chain reaction (PCR) has been widely used for single-nucleotide polymorphism (SNP) genotyping. However, the extraction of PCR-compatible genomic DNA from whole blood is usually required. This process is complicated and tedious, and is prone to cause cross-contamination between samples. To facilitate direct PCR amplification from whole blo...
